NR-14862 is a preparation of native MPT32/Apa (Gene Rv1860) purified from sterile-filtered Mycobacterium tuberculosis, strain H37Rv culture filtrate protein fraction. The culture filtrate protein fraction was precipitated with 40% ammonium sulfate, applied to a ConA column, eluted, concentrated, dialyzed, applied to a phenyl sepharose column and eluted. The purified MPT32/Apa was dialyzed in 10 mM ammonium bicarbonate.
Note: MPT32/Apa appears as a doublet by SDS-PAGE. The major component is 45 kDa and the minor component is a 42 kDa degradation product. The mature protein has a calculated molecular weight of 30.2 kDa, however the abundance of proline residues along with glycosylation causes this molecule to migrate at 45 kDa by SDS-PAGE.
Each vial contains approximately 500 ?g of lyophilized MPT32/Apa in 10 mM ammonium bicarbonate.
Note: NR-14862 is soluble in 100 mM to 500 mM aqueous buffered salt solutions, such as phosphate buffered saline. A 10 mM ammonium bicarbonate solution can also be used.
