NR-53528 is a preparation of periplasmic phosphate-binding lipoprotein, PstS1 (Gene Rv0934, non-acylated; also referred to as PhoS1), purified from the culture filtrate of Mycobacterium tuberculosis, strain H37Rv. The proteins soluble in 40% ammonium sulfate were precipitated by 70% saturated ammonium sulfate solution, suspended in ConA binding buffer and applied to a ConA column. ConA elution buffer, containing alpha-D-mannopyranoside, was used to elute the bound proteins. The bound material was applied to a phenyl sepharose column and eluted using a gradient of decreasing ammonium sulfate. Fractions containing pure 38kDa were pooled and dialyzed.
Note: The cell wall-associated and culture filtrate forms of PhoS1 seem to differ in
their hydrophobic characteristics. However, it has not been determined if there is a chemical difference between the cell wall and culture filtrate forms of the PhoS1 homologue.
Each vial contains approximately 100 ?g of lyophilized, purified NR-53528 in 10 mM ammonium bicarbonate.
Note: PstS1 is soluble in 100 mM to 500 mM aqueous buffered salt solutions, such as phosphate buffered saline. A 10 mM ammonium bicarbonate solution can also be used.
