NR-54970 (Vero E6-TMPRSS2-T2A-ACE2 cells; also referred to as Vero E6 cells expressing T2A ACE2 TMPRSS2 P2A) contains a preparation of Cercopithecus aethiops kidney epithelial cells (Vero E6; ATCC ? CRL-1586?) which were modified to express higher levels of transmembrane protease, serine 2 (TMPRSS2) and angiotensin-converting enzyme 2 (ACE2).
Vero E6-TMPRSS2-T2A-ACE2 cells were created in January 2021 by stably transfecting Vero E6 cells with plasmid (Addgene plasmid 41583; deposited by Phil Sharp, Massachusetts Institute of Technology) encoding ‘Puromycin-N-acetyltransferase-P2A-TMPRSS2-T2A-ACE2’ under chicken ?-actin promoter (CAG promoter). Puromycin-N-acetyltransferase confers resistance to puromycin, which is used to select transfected cells. P2A and T2A are short ‘self-cleaving’ peptides which enable expression of multiple proteins from a single mRNA. 2A-mediated self-cleavage occurs due to ribosome skipping the formation of a glycyl-prolyl peptide bond at the C-terminus of the 2A. P2A is from porcine teschovirus-1 and T2A is from Thosea asigna virus. ACE2 is a human receptor that is expressed widely, including in heart, kidney, small intestine and lung cells, and is involved in the regulation of hypertension and other cardiovascular diseases. The SARS-Related Coronavirus 2 (SARS-CoV-2) spike glycoprotein mediates viral binding to the host ACE2 receptor. TMPRSS2 has been shown to be involved in proteolytically activating SARS-CoV. Cells expressing human TMPRSS2 were found to be highly susceptible to infection by SARS-CoV-2.
Each vial contains approximately 1 mL of cell culture suspension frozen in Dulbecco’s Modified Eagle’s Medium containing 10% fetal bovine serum and 10 ?g per mL puromycin and DMSO (10%) cryopreservative. Sufficient cells are provided to initiate at least one new culture.
