NR-55500 Vero-hSLAM is a cell culture suspension of Cercopithecus aethiops kidney epithelial cells which were modified to express Human Signaling Lymphocytic Activation Molecule. Observations of working stocks of SARS-CoV-2 suggest that sequential propagation in Vero cells leads to critical changes in the region of the furin cleavage site (FCS). Global studies conducted at four institutions conclude that serial propagation of SARS-CoV-2 in Vero-hSLAM cells reduces the apparent rate of mutant production in stocks. Propagation in Vero-hSLAM appears to minimize mutations and improve the overall genetic stability of working stocks, as illustrated by Illumina deep sequencing of four sequential batches of stocks derived from an early clinical isolate propagated in Vero-hSLAM cells which found no detectable FCS deletions. Each vial contains approximately 1 mL of cell culture suspension frozen in Earle’s Modified Eagle’s Medium containing 10% fetal bovine serum and 0.4 milligrams per milliliter geneticin and DMSO (10%) cryopreservative. Sufficient cells are provided to initiate at least one new culture.
