NR-53821 contains a preparation of human (Homo sapiens) lung carcinoma epithelial cells (A549) expressing human angiotensin-converting enzyme 2 (ACE2). A549-hACE2 cells were created by stable insertion of human ACE2 driven by elongation factor-1a (EF-1a) promoter using the Sleeping Beauty transposon system pSBbi-BB (Addgene 60521). pSBbi-BB contains a blasticidin resistance gene for selection of transfected cells.

ACE2 is a human receptor that is expressed widely, including in heart, kidney, small intestine and lung cells, and is involved in the regulation of hypertension and other cardiovascular diseases. The SARS-Related Coronavirus 2 spike glycoprotein mediates viral binding to the host ACE2 receptor. This protein forms a trimer, and when bound to ACE2, allows fusion of the viral and cellular membranes, allowing viral entry and replication.

Each vial contains approximately 1 mL of cell culture suspension frozen in Dulbecco’s Modified Eagle’s Medium (69%) containing MEM non-essential amino acid solution (1%), fetal bovine serum (20%) and DMSO (10%) cryopreservative. Sufficient cells are provided to initiate at least one new culture.
