NR-55245 is a preparation of severe acute respiratory syndrome-related coronavirus 2 (SARS-CoV-2), isolate USA/CA_CDC_5574/2020 that has been inactivated by heating to 65°C for 30 minutes and diluted prior to vialing.
SARS-CoV-2, isolate USA/CA_CDC_5574/2020 was isolated from a nasopharyngeal swab on December 29, 2020 in San Diego County, California, USA.
Under the nomenclature system introduced by GISAID (Global Initiative on Sharing All Influenza Data), SARS-CoV-2, isolate USA/CA_CDC_5574/2020 is assigned lineage B.1.1.7 and GISAID clade GR using Phylogenetic Assignment of Named Global Outbreak LINeages (PANGOLIN) tool. The complete genome of SARS-CoV-2, isolate USA/CA_CDC_5574/2020 has been sequenced (GISAID: EPI_ISL_751801).
Intended Use:
NR-55245 is intended for use in molecular assays or can be used as a process (work-flow) control (for example: viral RNA extraction through qPCR amplification process). Please refer to the Certificate of Analysis for pre-inactivation infectious titer and post-inactivated genome equivalence (GE) quantification by Droplet Digital PCR for your specific lot. The GE is determined by full and partial genomes and is an estimate for end-users to qualify assays on their platform. The quantified products can also be used to make a dilution series to estimate level of detection. However, this product is not suitable as a whole cell antigen preparation because the protein content is largely contributed by the host cell and the fetal bovine serum used during virus propagation.
Inactivation Procedure:
Please refer to the Certificate of Analysis for the inactivation testing for the lot of material you have obtained from BEI Resources. In brief, 10% of the lot of inactivated virus is tested to confirm the absence of residual live virus by inoculating host cells. Using suitable controls, the test sample is passaged twice, with each passage incubated for two weeks. During procedure development, the inactivation protocol was validated by testing 100% of the sample of three independent lots for residual live virus. Note: The heat inactivation procedure is known to damage viral RNA and large fragments are not easily amplified. This material is suitable for qPCR assays for detection of small genome fragments.
Each vial contains approximately 0.25 mL of heat inactivated, clarified and diluted cell lysate and supernatant from Vero E6 cells infected with SARS-CoV-2, isolate USA/CA_CDC_5574/2020. The vial should be centrifuged prior to opening.
