Ricin toxin is a glycoprotein isolated from the seeds of Ricinus communis (R. communis), the castor bean plant. It is one of the most toxic and easily produced plant toxins. Ricin toxin consists of two polypeptide chains, A and B, linked by a disulfide bond. The A chain catalytically inactivates the eukaryotic 28S ribosomal RNA subunit, resulting in the inhibition of protein synthesis and death of the cell. The B chain is responsible for receptor binding and delivery of the toxin to the target cell. The sequence for the R. communis gene for the ricin toxin precursor protein has been reported (GenBank: X03179). The crystal structure for ricin toxin has been solved at 2.50 ? resolution (PDB: 2AAI). Ricin toxin has a molecular weight of approximately 63,000 daltons.
NR-720 reacts with a polyclonal antibody to recombinant ricin A chain as determined by radioimmunoassay. NR-720 is active in an in vitro cell-free protein synthesis inhibition assay, an in vitro cytotoxicity assay using human lymphoma cells and an in vivo toxicity assay in mice.
Each vial of NR-720 contains approximately 5 mg of ricin toxin suspended in 0.2 M sodium acetate buffer, pH 3.5.
