The blasticidin-resistance gene (BSD) was amplified from the pcDNA6/V5-His plasmid (Invitrogen) using the primer pair 5'-GGAAGATGCATGCCAAGCCTTTGTCTCAAGAATCC ACCCTC-3' and 5'-GACGGGAAGCTTTGCTCCTCGGCCACGAAGTGC-3'. Amplified inserts were digested with NsiI and HindIII and used to replace the Toxoplasma gondii DHFR-TS in the pDT-Tg23 vector, yielding construct pCBM-BSD. The resulting size of the plasmid is 5.6 kilobases. The sequence has not been confirmed by BEI Resources. pCBM-BSD transfected P. falciparum 3D7 parasites confer resistance to blasticidin.Each vial of MRA-103 contains approximately 20 ?L of purified plasmid DNA in buffer at a concentration of 100 ng/20 ?L.Mamoun, C. The pSEL1 vector was digested with HindIII and a fragment ligated that was generated by annealing two oligonucleotides to form a NotI, SmaI, XhoI and HindIII polylinker (pSEL1-NSXH). The HRP3 5' sequence was obtained by PCR using as a template the plasmid pDT.Tg23 and oligonucleotides 10F and 10R (HRP3 5'). The 1.6 kb fragment was digested with HindIII and XhoI and subcloned directly into pSEL1-NSXH digested with HindIII and XhoI. This resulted in the generation of pHC5.
