The pSEL1 vector was digested with HindIII and a fragment ligated that was generated by annealing two oligonucleotides to form a NotI, SmaI, XhoI and HindIII polylinker (pSEL1-NSXH). The HRP3 5' sequence was obtained by PCR using as a template the plasmid pDT.Tg23 and oligonucleotides 10F and 10R (HRP3 5'). The 1.6 kb fragment was digested with HindIII and XhoI and subcloned directly into pSEL1-NSXH digested with HindIII and XhoI. This resulted in the generation of pHC5. The resulting size of the plasmid is 8.634 kilobases.Each vial of MRA-12 contains approximately 1 ?g of purified plasmid DNA in buffer at a concentration of 1 ?g/20 ?L.Crabb, B. S., et al.
